Effect of Liver injury, Auto-And Homotransplantation on Liver Nucleic Acid Content And transaminase Activity


Abstract
One hundred and ten rabbits were divided into 5 groups as follows :
Group 1 : Control (Sham operated)
Group 2 : Resected at 1/3 portion of left lobe of the liver.
Group 3 : Resected at 1/3 portion of left lobe of the liver and autotranplanted in right inguinal region.
Group 4 : Ligated at 1/3 portion of left lobe of the liver.
Group 5 : Sham operated and homotransplanted in right inguinal region.
Fourteen out of 22 rabbits of each group were killed at 24 hours after operation, 8 out of them of each group were killed at 48 hours after operation. Immediately after operation, all animals were given 301gc of Pat intraperitoneally. After killing the animals, middle lobe of the liver were taken out to determine nucleic acid content, P32 incorporation rate into liver nucleic acid and transaminase activity. The results of this experiment were as follows;
(1) A. RNA-P content of the liver at 24 hours after operation was 35.3 mg-p/100 gm tissue in the 1st group, 45.2(125.2%) in the 2nd group, 43.6 in the 3rd group, 40.9 in the 4th group and 32.4 in the 5th group. The quantitative order was 2nd group> 3rd group> 4th ,:Youp>1st group> 5th group.
B. RNA-P content of the liver at 48 hours after operation was 34. lmg--p/100 gm tissue in the 1st group,41.8(122.2%) in the 2nd group,40.5 in the 3rd group,41.1 in the 4th group and 28.1 in the 5th group. The quantitative order was 2nd gr.> 4th gr.> 3rd gr. 1st gr.> 5th gr.
(1l) A. DNA-P content of the ¢¥liver at 24 hours after operation:was z19;1:fng-P/100 gmm tissue in the 1st group, :30.5(159.7%) in the 2nd group, 28.5 in the 3rd group,26.7 in the 4th group and 16.5 in the 5th group The `quantitative order was 2nd ,gr.> 3rd gr.> 4th gr.) 1st gr.> 5th gr.
B. DNA-.P content of the liver at 48 hours after operation was 20.5 mg P/100 gm tissue In the 1st group, 28.2(137.6%) in the 2nd group, 28.5 in the 3rd group, 26.8 in the 4th group .and 18.3 in the 5th group. The quantitative order was 3rd -gr.) 2nd gr. )-4th gr.> 1st gr.> 5th gr..
(1f) A. GPT activity of the liver at 24 hours after operation was?36.900 units/gm tissue in the 1st group, 44.550(120.7%) in the 2nd group, 43.080 in the 3rd group, 43.900 in the 4th group and 37.800 in the 5th group. The quantitative order was 2nd gr. )4th gr. ) 3rd gr.) 5th gr.) 1st gr.
B. GPT activity of the liver at 48 hours after operation was 45.370 units/gm tissue in the 1st group, 62.850 in the 2nd group, 59.000 in the 3rd group, 84.450(186.4%) in the 4th group and 48.600 in the 5th group. The quantitative order was 4th gr. 2nd gr.) 3rdgr.) 5th gr.> 1st gr.
(If) A. GOT activity of the liver at 24 hours after operation was 102.350 units/gm tissue in the 1st group, 129,990 in the 2nd group, 153,450(149.9%) in the 3rd group, 107.200 in the 4th group and 106.500 in the 5th group. The quantitative order was 3rd gr.> 2nd gr. ) 4th gr.) 5th gr.> 1st gr.
B. GOT activity of the liver at 48 hours after operation was 128.000 units/gm tissue in the 1st group, 151.350 in the 2nd group, 139.500 in the 3rd group, 186.900(146%) in the 4th group and 136.900 in the 5th group. The quantitative order was 4th gr.> 2nd gr.> 3rd gr.> 5th gr. )1st gr.
(V) A. The specific activity of P32 incorporation rate into RNA fraction of the liver at 24 hours alter operation was 3154.8 epm/mg-.P in the 1st group, 5677.4(180%) in the 2nd group, 5526.6 in the 3rd group, 5674.3 in the 4th group and 2972.8 in the 5th group. The quantitative order was 2nd gr.>4th gr.) 3rd gr.> 1st gr.> 5th gr.
B. The specific activity of P32 incorporation rate into RNA fracticn of the liver at 48hours after operation was 2187.0 cpm/mg-P in the 1st group, 4609.2(210.8%) in the 2nd group, 3919.4 in the 3rd group, 3122.7 in the 4th group and 2710.3 in the 5th group. The quantitative order was 2nd gr.) 3rd gr.) 4th gr.> 5th gr.) 1st gr.
(II) A. The specific activity of P32 incorporation rate into DNA fraction of the liver . at 24 hours after operation was 1570.7 cpm/mg-P in the 1st group, 2645.9(168.5%) in the 2nd group, 2549.6 in the 3rd group, 2065.5 in the 4th group and 1512.1 in the 5th group. The quantitative order was 2nd gr.) 3rd gr.> 4th gr.) 1st gr.> 5th gr.
13. The specific activity of P32 incorporation rate into DNA fraction of the liver at 48 hours after operation was 1458.5 cpm/mg-P in the 1st group, 2806.7(192.4%) in the 2nd group, 2247.4 in the 3rd group, 2065.3 in the 4th group. and 1278.6 in the 5th group. The quantitative order was 2nd gr.> 3rd gr.> 4th gr.> 1st gr.> 5th gr.
From the results thus obtained, it may be concluded that the liver injury causes prompt increase of liver nucleic acid content and tranaminase activity, and that transplanted liver tissue inhibits this regenerative reaction.
It may be also suggested that increased activity of serum transaminase of liver injured animals resulted from compensatory increase in enzyme production by healthy liver tissue and the source of increased enzyme is healthy, not necrotic, tissue.
